What is pyridoxal 5'-phosphate (P-5-P)?

Pyridoxal 5'-phosphate (P-5-P) is the active coenzyme form of vitamin B6 that is directly utilized by the body. Vitamin B6 naturally occurs in 3 major chemical forms: pyridoxine, pyridoxal, and pyridoxamine. While each of these forms has vitamin activity, only P-5-P (also called PLP) has an active coenzyme function in the body. P-5-P plays an important role in amino acid metabolism, neurotransmitter production, and a variety of other metabolic reactions. Non-P-5-P forms of vitamin B6 are generally converted to P-5-P before being utilized in the body.

How does P-5-P compare to other forms of B6 such as pyridoxine hydrochloride?

Although pyridoxine hydrochloride is the most commonly used form of vitamin B6 in dietary supplements, and the best-studied, it must be converted to the active coenzyme form, P-5-P, to be utilized by the body. In certain clinical settings, subjects responded better to P-5-P than to pyridoxine, suggesting that they had less ability to convert pyridoxine to P-5-P. In contrast, vitamin B6 as P-5-P is already fully "active" and readily usable, as it does not require further transformation in the body.

Is there any scientific documentation that supports enhanced bioavailability of P-5-P over pyridoxine hydrochloride?

Currently, little research exists comparing the efficacy of pyridoxal 5'-phosphate with other forms of vitamin B6 such as pyridoxine. In certain clinical settings, subjects responded better to P-5-P than to pyridoxine, suggesting that they had less ability to convert pyridoxine to P-5-P. Although further research is certainly warranted, available research studies suggest P-5-P may be an optimal way to support vitamin B6 levels and actions in the body, in particular for individuals who cannot convert pyridoxine, such as those requiring the vitamin for B6-dependent red blood cell formation, B6-dependent neurological function, and individuals on the spectrum.

Why is magnesium added to the P-5-P Plus formulation?

Magnesium is included to assist vitamin B6 metabolism. Metabolic functions such as amino acid metabolism and neurotransmitter production require both magnesium and P-5-P. In addition, these 2 nutrients are frequently found as cofactors for the same enzymes.
